Kisspeptin-10 (also known as metastin fragment or Kp-10) is a synthetic 10-amino acid peptide (Tyr-Asn-Trp-Asn-Ser-Phe-Gly-Leu-Arg-Phe-NH₂) derived from the C-terminal region of the KISS1 gene product. It acts as the primary endogenous ligand for the kisspeptin receptor (KISS1R/GPR54), a G-protein-coupled receptor highly expressed on GnRH neurons in the hypothalamus.

By binding KISS1R, Kisspeptin-10 potently stimulates gonadotropin-releasing hormone (GnRH) secretion, which in turn drives rapid and robust release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) from the anterior pituitary. This upstream activation of the hypothalamic-pituitary-gonadal (HPG) axis makes Kisspeptin-10 one of the most powerful known stimulators of endogenous gonadotropin secretion. Human and animal studies demonstrate that Kisspeptin-10 elicits dose-dependent increases in LH (often 8–15-fold or higher) and FSH, with significant downstream elevations in testosterone in men and sex steroid production in women. It exhibits sexual dimorphism, with particularly strong gonadotropin responses in men and during specific phases of the menstrual cycle in women. Kisspeptin-10 also influences central pathways involved in sexual behavior, arousal, motivation, and mood regulation.

In research and hormone optimization contexts, Kisspeptin-10 is administered via subcutaneous or intravenous injection (typical research doses 100–200 mcg per bolus, or lower for repeated pulsatile protocols) to explore HPG axis function, fertility restoration, hypogonadotropic hypogonadism, delayed puberty, hypoactive sexual desire disorder (HSDD), and natural testosterone enhancement without exogenous hormone suppression. Many users report increased libido, improved sexual function, better mood, and enhanced energy when used in short cycles. While early clinical studies show promising effects on gonadotropin and sex steroid levels with good tolerability (mild transient flushing, headache, or nausea), Kisspeptin-10 is not FDA-approved for any therapeutic, fertility, hormone optimization, libido enhancement, or medical use as of 2026—it remains strictly investigational.

No large-scale, long-term human trials have established definitive safety or efficacy for systemic use. Potential risks include pituitary overstimulation with continuous administration (possible receptor desensitization), transient hormone fluctuations, or unknown long-term effects on reproductive tissues. Kisspeptin-10 should only be used under strict medical supervision with hormone monitoring (LH, FSH, testosterone, estradiol) and proper aseptic technique. It is prohibited by WADA for athletes and should never be used without guidance from a qualified healthcare provider experienced in reproductive endocrinology or peptide research.
